Collette Wolfe made her film debut in 2006 when she appeared in the film ‘The Foot Fist Way’. This was followed by a role in 'Great World of Sound' that garnered the Grand Jury Award at that year’s Atlanta Film Festival. Then in 2008, she did the movies ‘Semi-Pro’ and ‘Four Christmases’. While the former failed to earn any profit at the American box office, the latter became a hit, eventually earning $163.7 million worldwide. Her roles in 2009 included guest appearances in the drama series ‘Reaper’ and ‘Greek,’ and supporting roles in the comedy movies ‘17 Again’ and ‘Observe and Report’.

During 2010 and 2011, Wolfe played Kirsten in the sitcom ‘Cougar Town’. She also featured in the movies ‘Hot Tub Time Machine’ and ‘Young Adult’ during this period.

Soon after, she had a supporting role in Jay Chandrasekhar's ‘The Babymakers’ (2012), a comedy flick about a man, who, after suspecting he is infertile, tries to rob a sperm bank. Also in 2012, the actress featured in the television drama ‘Fetching’ as Liza.

The year 2013 saw Wolfe in guest appearances in many TV shows, including ‘The Office’ and ‘Mad Men’. That year, she also filmed four episodes of the mid-season replacement ‘Next Caller’ that never aired. This was followed by her appearance in the science fiction film 'Interstellar’. The storyline follows a group of astronauts who travel all the way through a wormhole near Saturn in the hunt for a new home for humanity.

From 2015 to 2016, Wolfe played the role of Dorothy Durwood in the comedy-drama TV series ‘You're the Worst’. During this time, she also portrayed the character of Shawni in the sitcom ‘Your Family or Mine’. In 2018, she had a guest role in an episode of ‘Grey’s Anatomy’ as well as a recurring role in the comedy series ‘A.P. Bio’.
